Abstrak:
Tujuan penelitian ini adalah membandingkan ketelitian dan ketepatan metoda titrasi (kupri sulfat sebagai standar) dengan metoda spektrofotometri (amonium molibdat sebagai pereaksi pembentuk warna) pada penentuan asam askorbat dalam buah pepaya.
Tujuan penelitian ini adalah membandingkan ketelitian dan ketepatan metoda titrasi (kupri sulfat sebagai standar) dengan metoda spektrofotometri (amonium molibdat sebagai pereaksi pembentuk warna) pada penentuan asam askorbat dalam buah pepaya.
Pada penelitian ini diketemukan
bahwa kondisi optimal metoda titrasi dengan kupri sulfat 0,01 M sebagai larutan
standar untuk penentuan asam askorbat diperoleh pada penambahan 1 g kalium
iodida (= 10 mL larutan XI 10%) dan 5 tetes larutan 1% kanji (amilum),
sedangkan jumlah asam askorbat yang dianalisa sebaiknya antara 0,5 mg sampai 10
mg per 5 mL, larutan. Selanjutnya kondisi optimal metoda spektrofotometri di
daerah tampak dengan pereaksi amonium molibdat untuk penentuan asam askorbat
diperoleh pada penambahan pereaksi asam metafosfat-asam asetat 0,5 mL, larutan
asam sulfat 5% 4 mL, larutan amonium molibdat 5% 2 mL dan absorbans diukur pada
panjang gelombang 700 nm. Warna kompleks biru molibden yang terbentuk antara
asam askorbat dan amonium molibdat stabil dalam waktu satu jam. Hukum
lambert-Beer terpenuhi sampai konsentrasi 56 ppm, tetapi pengukuran absorbans
sebaiknya dilakukan pada range konsentrasi 10 sampai 40 ppm. Besi (II) dan besi
(III) tidak mengganggu sampai jumlah maksimum 25Mg.
Kondisi-kondisi optimal yang
ditemukan dari ke dua metoda tersebut digunakan pada penentuan asam askorbat
dalam buah pepaya. Dengan menggunakan metoda statistik yang sesuai, ditemukan
bahwa simpangan baku (S) metoda titrasi (S = 1,65) lebih tinggi dari simpangan
baku metoda spektrofotometri (S = 0,45). Persen perolehan kembali asam askorbat
dengan menggunakan metoda titrasi adalah 95,0%, sedangkan persen perolehan
kembali asam askorbat dengan menggunakan metoda spektrofotometri adalah 98,4%.
Jadi dapat disimpulkan bahwa untuk penentuan asam askorbat dalam buah pepaya,
ketelitian dan ketepatan metoda spektrofotometri di daerah tampak dengan
menggunakan amonium molibdat sebagai pereaksi pembentuk warna lebih baik dari
pada ketelitian dan ketepatan metoda titrimetri dengan menggunakan larutan
standar kupri sulfat.

The aim of this investigation is to compare the precision and accuracy of the determination of ascorbic acid in papaya fruit by a titration method using copper sulphate as a standard and by a spectrophotometric method using ammonium molybdate as a colour forming agent.
Abstract:
The aim of this investigation is to compare the precision and accuracy of the determination of ascorbic acid in papaya fruit by a titration method using copper sulphate as a standard and by a spectrophotometric method using ammonium molybdate as a colour forming agent.
It is found in this investigation
that the optimal conditions for the titration method using 0,01 M copper
sulphate as a standard solution were obtained by adding 1 g of potassium iodide
(= 10 mL 10% XI solution) and 5 drops of a 1% starch (amylum) solution, whilst
the amount of ascorbic acid analyzed should be between 0,5 mg and 10 mg per 5
mL of solution. Optimal conditions for the spectrophotometric method in the
visible region using ammonium molybdate reagent were obtained by adding 0,5 mL
of metaphosphoric acid-acetic acid reagent, 4 mL of a 5% sulphuric acid
solution and 2 mL of a 5% solution of ammonium molybdate and measuring the
absorbance at a wavelength of 700 nm. The colour of the molybdenum blue complex
formed between ascorbic acid and ammonium molybdate was stable for 1 hour. The
Lambert-Beer Law is obeyed up to a concentration of 56 ppm, but absorbance
measurements are best done within the concentration range of 10 to 40 ppm. Iron
(II) and iron (III) do not interfere up to a maximum amount of 25 Mg.
The optimum conditions found were
employed in carrying out the ascorbic acid determination in pepaya fruit using
both methods. By using the appropriate statistical methods, it was found that
the standard deviation (S) of the titration method (S = 1,65) is higher than
the standard deviation of the spectrophotometric method (S = 0,45). The percent
recovery of ascorbic acid using the titration method is 95,0%, whilst the
percent recovery using the spectrophotometric method is 98,4%. It may be
concluded that for the determination of ascorbic acid in pepaya fruit, the
precision and accuracy of the visible region spectrophotometric method
employing ammonium molybdate as a colour forming reagent is better than the
precision and accuracy of the titrimetric method using copper sulphate standard
solution.
